Flt3-ligand (FL) is a recently identified hematopoietic cytokine whose activities are mediated by binding to the transmembrane glycoprotein Flt3. Flt3 was first discovered as a member of the class III subfamily of receptor tyrosine kinases (RTK) whose expression among hematopoietic cells was found to be restricted to highly enriched stem/progenitor cell populations. Additionally, class III RTKs include the receptors from SCF, M-CSF and PDGF. Not surprisingly, Flt3-ligand is also structurally related to M-CSF and SCF. All three cytokines have been shown to exist both as type I transmembrane proteins and as soluble proteins. The predominant human FL isoform is a transmembrane protein that can undergo proteolytic cleavage to generate a soluble form of the protein. An alternatively-spliced FL mRNA, encoding a soluble form of the human FL, has also been identified. FL is widely expressed in various human and mouse tissues. At the amino acid sequence level, human and mouse FL are approximately 72 % identical and the two proteins exhibit cross-species activity. FL has been shown to synergize with a wide variety of hematopoietic cytokines to stimulate the growth and differentiation of early hematopoietic progenitors.

Tumor necrosis factor alpha (TNF-alpha), also called cachectin, is the best-know member of the TNF-family, which can cause cell death. This protein is produced by neutrophils, activated lymphocytes, macrophages, NK cells, LAK cells, astrocytes endothelial cells, smooth muscle cells and some transformed cells. TNF-alpha occurs as a secreted, soluble form and as a membrane-anchored form, both of which are biologically active. The naturally-occurring form of TNF-alpha is glycosylated, but non-glycosylated recombinant TNF-alpha has comparable biological activity. The biologically active native form of TNF-alpha is reportedly a trimer. Human and murine TNF-alpha show approximately 79 % homology at the amino acid level and cross-reactivity between the two species. Two types of receptors for TNF-alpha have been described and virtually all cell types studied show the presence of one or both of these receptor types.

Granulocyte-Macrophage Colony Stimulating Factor (GM-CSF) is secreted by a number of different cell types (including activated T cells, B cells, macrophages, mast cells, endothelial cells and fibroblasts) in response to cytokine or immune and inflammatory stimulation. It was initially characterized as a growth factor that can support the in vitro colony formation of granulocyte-macrophage progenitors and has functions of stimulates the growth and differentiation of hematopoietic precursor cells from various lineages. GM-CSF has also been reported to have a functional role on non-hematopoietic cells and can induce human endothelial cells to migrate and proliferate. Additionally, it can stimulate the proliferation of a number of tumor cell lines, including osteogenic sarcoma, carcinoma and adenocarcinoma cell lines. Human GM-CSF shares 54 % sequences identity with mouse GM-CSF, but has no biological effects across species. GM-CSF is used as a medication to stimulate the production of white blood cells following chemotherapy and has also recently been evaluated in clinical trials for its potential as a vaccine adjuvant in HIV-infected patients.

Human Interleukin-15 (IL-15) is expressed by the IL15 gene located on the chromosome 4. It shares approximately 97 % and 73 % sequence identity with simian and murine IL-15, respectively. Both human and simian IL-15 are active on murine cells. IL-15 is secreted by mononuclear phagocytes (and some other cells), especially macrophages following infection by virus. It possesses a variety of biological functions, including stimulating and maintaining of cellular immune responses, especially regulating T and natural killer (NK) cell activation and proliferation. In additionally, it shares many biological properties with IL-2, including T, B and NK cell-stimulatory activities. IL-15 signals through a complex composed of IL-2/IL-15 receptor beta chain. Although IL-15 lacks sequence homology with IL-2, it has recently been shown that both the beta and gamma chains of the IL-2 receptor are utilized for IL-15 binding and signaling. In addition, an IL-15 specific binding protein has also been cloned from a mouse T cell clone.

Interleukin-7 (IL-7) is encoded by the IL7 gene and secreted by stromal cells in the red marrow and thymus. It binds to the IL-7 receptor, a heterodimer consisting of IL-7 receptor alpha and IL-2 receptor gamma chain. IL-7 stimulates the differentiation of hematopoietic stem cells into lymphoid progenitor cells and also stimulates proliferation of B cells, T cells and NK cells. IL-2 is a powerful immunoregulatory lymphokine produced by T-cells in response to antigenic or mitogenic stimulation. It is expressed by CD4+ and CD8+ T cells, gammadelta T cells, B cells, dendritic cells, and eosinophils. IL-2/IL-2R signaling is required for T-cell proliferation and other fundamental functions which are essential for the immune response. The receptor for IL-2 consists of three subunits (55 kDa IL2Ralpha, 75 kDa IL2Rβ, 64 kDa common gamma chain gammac/IL2Rgamma) that are present on the cell surface in varying preformed complexes. Mature human IL-2 shares 56 % and 66 % amino acid sequence identity with mouse and rat IL-2, respectively. Human and mouse IL-2 exhibit cross-species activity.

We recommend that this vial be briefly centrifuged prior to opening to bring the contents to the bottom. Reconstitute in sterile distilled H2O to a concentration of 0.1-1.0 mg/mL. Stock solutions should be apportioned into working aliquots and stored at ≤ -20 °C. Further dilutions should be made in appropriate buffered solutions. Do not reconstitute in cell culture media directly.

Interleukin-1 alpha (IL-1alpha) is a non-secreted proinflammatory cytokine produced mainly by activated macrophages, as well as neutrophils, epithelial cells, and endothelial cells. It possesses metabolic, physiological, haematopoietic activities, and plays one of the central roles in the regulation of the immune responses. Both IL-1alpha and IL-1β binds to the same receptor and has similar identical biological properties. Among various species, the amino acid sequence of mature IL-1alpha is conserved 60 % to 70 % and human IL-1 has been found to be biologically active on murine cell lines. IL-1alpha recently started to find effective application in cosmetic and dermatological formulations, which allow to significantly harmonizing derma architecture.
